The impact of climate change on naturally occuring Cholera species is of great interest to US Biodefense and Artificial Intelligence is being applied.2345

As previously mentioned7 the US Government reported its work on Bioweapons to the UN in 2019 in its “Confidence Building” document with much of the effort at the U.S. Army Medical Research Institute of Infectious Diseases (USAMRIID), Fort Detrick, Frederick, Maryland.

USAMRIID had a budget of just under US$100,000,000 1 October 2017 to 30 September 2018.

In that document they reveal that they collaborated with Cholera toxin researchers in Marburg Germany, Manchester UK and The Tauri Group in Virginia to look at chromosome transfer during cell division.8

Abstract

Chromosomal inheritance in bacteria usually entails bidirectional replication of a single chromosome from a single origin into two copies and subsequent partitioning of one copy each into daughter cells upon cell division.

However, the human pathogen Vibrio cholerae and other Vibrionaceae harbor two chromosomes, a large Chr1 and a small Chr2.

Chr1 and Chr2 have different origins, an oriC-type origin and a P1 plasmid-type origin, respectively, driving the replication of respective chromosomes.

Recently, we described naturally occurring exceptions to the two-chromosome rule of Vibrionaceae: i.e., Chr1 and Chr2 fused single chromosome V. cholerae strains, NSCV1 and NSCV2, in which both origins of replication are present.

Using NSCV1 and NSCV2, here we tested whether two types of origins of replication can function simultaneously on the same chromosome or one or the other origin is silenced.

We found that in NSCV1, both origins are active whereas in NSCV2 ori2 is silenced despite the fact that it is functional in an isolated context.

The ori2 activity appears to be primarily determined by the copy number of the triggering site, crtS, which in turn is determined by its location with respect to ori1 and ori2 on the fused chromosome.

Cholera in Democratic Republic of Congo

A very recent paper looked at modeling Climate-Driven Cholera in Democratic Republic of Congo and Nigeria.9

Very relevant considering the current Ebola disaster in Congo and surrounding countries with risk of export beyond Africa.

Other mentions of Cholera and its Toxins

I have touched on Cholera before, including the work of Jane Yeadon in attempts to develop a Cholera Jab.10

Cholera Jabs had a Death to report ration of 5,524 per Million when I looked two years ago.11

Curtin University in Western Australia includes Cholera in its List of Weaponisable Pathogens or Toxins.12

Elisabeth Bik was involved in developing Jabs for new strains of Vibrio cholerae involved in Cholera epidemics across India and Bangladesh.13

The organism causing Cholera is closely related to the E. coli bacteria used in production of Covid19 and other Jabs.14

Akiko Iwasaki and coworkers performed detailed research in 2009 showing Cholera toxin inhibits IL-12 production and CD8α⁺ dendritic cell differentiation by cAMP-mediated inhibition of IRF8 function.15

Cholera Toxins used as “Adjuvants” in Jabs

That’s right, many Jabs under development use the Cholera poisons to shock your system.

An example back in 1990.16

I mentioned17 one example from a Curevac (now wholly owned subsidiary of BioNTech) patent that included these lovelies:

Cholera Toxin (CT), Cholera holotoxin, mCT-E1 12K, Cholera Toxin B subunit (CTB), Cholera Toxin A1- subunit-ProteinA D-fragment fusion protein, CTA1-DD gene fusion protein, Chimeric A1 subunit of Cholera Toxin (CTA1 )-DD, E. coli heat-labile Enterotoxin (LT), LT(R192G), LTK63, LTK72, LT-R192G, LT B subunit, LT-OA (E coli labile enterotoxin protoxin).

I also pointed out the some Cholera enthusiasts want to squirt Cholera Toxin B up your nose.18

Mark Zuckerberg patent19 says:

“Other exemplary adjuvants include, but are not limited to, Cholera toxin ..”.

In 2021 Moderna published a patent claiming they could make a Jab against Hanta virus and they included Cholera toxin, Cholera toxin B subunit among their list of “adjuvants”.20

PubMed lists 15,296 peer-reviewed papers when seaching “Cholera toxin”.

Of those 1,783 papers mention the structure.

Glycocalyx and Cholera

Recently Martha Carlin and her Australian coworkers Barry Ninham and Matthew Battye published an important review, including 116 references, on the emerging understanding of the role of the Glycocalyx in numerous diseases, with a focus on Parkinsons disease.21

Their work primed me to see a recent article on the Glycocalyx related to Cholera by Laia Saltor Núñez et al.22

ABSTRACT:

AB5 toxins are a class of bacterial toxins that recognize cell surface carbohydrates to facilitate their uptake by the target cell.

Among them are Cholera Toxin (CT) from Vibrio cholerae that causes Cholera, and Shiga toxin (STx) from Shigella dysenteriae and certain strains of Escherichia coli, which cause Hemolytic Uremic Syndrome.

While the glycolipid ligands for CT and STx (gangliosides GM1 and Gb3, respectively) have long been known, recent studies have shown that fucosylated structures, like Lewisx (Lex), also play a role in CT binding.

This realization raises questions about the importance of interactions between these toxins and nonglycolipid components of the Glycocalyx, which are not well understood.

To address this challenge, we created Glycocalyx models of defined thickness and tunable molecular composition through grafting of mucin-like glycopolymers on solid-supported lipid bilayers (SLBs).

The synthesized mucin-like glycopolymers comprised a Hyaluronic Acid (HA) backbone, an anchor tag (biotin or hexa-histidine) at the HA reducing end, and side chains of relevant oligosaccharides (Lex, Gb3, or lactose) at defined densities. Analyses by quartz crystal microbalance with dissipation monitoring and spectroscopic ellipsometry provided quantification of the thickness, mesh size, and target glycan concentration of the Glycocalyx models and of toxin binding kinetics. The B subunit pentamers of both CT and STx showed significantly enhanced affinity in the model Glycocalyx environment due to multivalent binding to their respective target glycans.

Most notably, toxin binding increased superlinearly with the concentration of the target glycan in the model Glycocalyx.

We propose that such “superselective” binding is an important factor in host cell selection. Our approach provides a new set of tools to make designer Glycocalyces and analyze multivalent protein-glycan interactions in a controlled environment.

Recall that Pfizer reported Deaths from Sepsis and specifically Shigella in its Covd19 Jab trials.23

Today I will briefly look at the Nebraska born Ebola expert, Alan Lee Schmaljohn,1 husband of Connie (Née Hrdlicka) the Hantavirus expert mentioned recently.2

Alan Schmaljohn publishes an impressive c.v. and he was Working Group Leader for Filovirus vaccines at U.S. Army Medical Research Institute of Infectious Diseases (USAMRIID), located on Fort Detrick, Maryland, until his voluntary retirement from government service in 2007.

Thanks to former US Bioweapons Lab Inspector Meryl Nass, who pointed me toward him as she knows the couple.

Alan Schmaljohn has 52 papers listed on PubMed from 1982 to 2019.

He also has a number of patents for Ebola that could take you quite a long time to read and fully appreciate. I have only just begun.

After a period of translational research at UMB with an Ebola vaccine that evolved from his prior work, Dr. Schmaljohn has turned more recently to a part-time role that involves teaching, writing, advising, and service—while resigning from personally directed laboratory research.

Both Alan and Connie are interested in Virus Reassortment.3

Alan Schmaljohn’s work includes discovery of how Ebola replicates in monocyte-derived dendritic cells without inducing the production of cytokines and full maturation. He knows a lot about Endotoxin.

He identifed Folate receptor-alpha as a cofactor for cellular entry by Ebola viruses.

He has patented creation of Virus-like particles for use in Jabs.

Trump and RFK Jr. appointment, Dr Steven Hatfill called Anthony Fauci a liar in 2021 as the mRNA Covid19 Jab Horror was becoming apparent. He investigated Lethal Blood Clotting and Haemorrhage and used various laboratory animals to investigate species variation in Virus Lethality while working at Fort Detrick.4

Last time I looked at US VAERS database Ebola Jabs had 19,607 Deaths per Million reports, suggesting a combination of ineffectiveness and outright toxicity.7

John Su identified risk of using Live Viruses in Ervebo Ebola Jabs.8

Tromethamine is also used in the Ervebo (Ebola Zaire live virus) Jab.9

Moncef Slaoui, who has a research centre in the heart of the US Bioweapons state of Maryland named after him at GSK Rockville, has worked, with special agreement with WHO, on development of Jabs including Ebola.10

Mark Zuckerberg and his mates at Chan Zuckerberg Biohub in San Francisco (CZ Biohub SF) have a ceased patent application for Ebola Virus Jabs.11

Ebola is one of numerous viruses that exploit our Furin to assist with cell entry.12

Ebola has dozens of entries in the US report on BioWeapons “Confidence Building” that I mentioned before when discussing Israeli research into another Bioweapon.13

Let’s look at two major studies where Person-to-Person transmission occurred with high Fatality rate caused by the Andes variant of Hantavirus.

Hantavirus Deaths in 1996-1997

In my earlier article3 I referenced the elegant paper4 by Padula et al. published in 1998 looking at a mass Death outbreak of the Andes variant of Hantavirus from September 1996 to January 1997 in El Bolsón, Argentina.

The authors used highly reliable PCR and the best available Genomic Sequencing to track contacts from patients zero.

Abstract

An increase of Hantavirus Pulmonary Syndrome (HPS) cases around a southwestern Argentina town and in persons living 1400 km away but in contact with those cases was detected during the spring of 1996.

In order to evaluate person-to-person transmission we compared the homology of PCR-amplified viral sequences of 26 Argentine and Chilean cases.

Sixteen of them were epidemiologically linked cases and had the same sequence (Epilink/96) in the S segment 3′ noncoding region and in the M segment partial G1 and G2 region (a total of 1075 nucleotides).

Contrarily, two geographical and contemporary but nonepidemiologically related cases differed from Epilink/96 in the compared regions.

No significant differences, such as glycosylation or hydrophilic pattern, were found between Epilink/96 and the other sequences.

Nucleotide and deduced amino acid sequence homologies between samples from southern Argentina and Chile ranged from 90.9 to 100% and 96.4 to 100%, respectively.

Phylogenetic analysis revealed that all the analyzed southwestern viruses belong to the Andes lineage.

Although human infection principally occurs via inhalation of contaminated rodent excreta, our results with Andes virus show the first direct genetic evidence of Person-to-Person Transmission of a Hantavirus.”

Solid State 100% ELISA Test Kit

Padula and coworkers devised a reliable diagnostic test for their serum analysis of people with suspected Andes variant of Hantavirus.5

Abstract

Hantavirus Pulmonary Syndrome (HPS) with high mortality rate has been reported in five countries in South America.

Rapid accurate methods are important both for monitoring acute infections and for epidemiological studies.

The Andes virus nucleoprotein amino acid sequence has a high identity percentage compared with other sequences of this region and has been chosen for the development of diagnostic reagents.

Andes nucleoprotein expressed in Escherichia coli was applied as antigen in IgG, IgA and mu-capture IgM enzyme-linked inmunosorbent assays (ELISAs).

An evaluation of this reagent was conducted to establish its usefulness for differential diagnosis of HPS and seroprevalence studies. Samples from 135 reverse transcription (RT)-PCR-confirmed HPS cases, 77 individuals with other respiratory infections and 957 healthy inhabitants from endemic and non-endemic areas were analysed.

The Hantavirus-infected patients had an early and strong IgM, IgG and IgA serum antibody response, in most of the cases as early as 1, 7 and 1 days following onset of symptoms, respectively.

IgM and IgG detection showed a specificity and sensitivity of 100%.

Andes-specific IgM antibodies were found in all patients in the first available sample, which remained detectable for at least 43 days.

Specific IgA antibodies were also detected in Saliva of patients with acute HPS.

The short duration of the disease and the risk for contacts due to person-to-person transmission of Andes virus necessitate the use of highly sensitive tests which might lead to earlier detection of infected people and improve the treatment and management of patients with HPS.

Hantavirus Deaths from 2018 Superspreader Party

Another very interesting Person-to-Person spreading Andes Hantavirus paper worth a read by Martínez and 31 other authors led by U.S. Army Medical Research Institute of Infectious Diseases, Fort Detrick, Frederick - famous for its holding stocks of Bioweapons Grade Viruses and GMO manipulation of them.6

They looked at Epuyén cases and compared samples from a number of other Andes Hantavirus Fatal outbreaks.

RESULTS

After a single introduction of ANDV from a rodent reservoir into the human population, transmission was driven by 3 symptomatic persons who attended crowded social events.

After 18 cases were confirmed, public health officials enforced isolation of persons with confirmed cases and self-quarantine of possible contacts; these measures most likely curtailed further spread.

The median reproductive number (the number of secondary cases caused by an infected person during the infectious period) was 2.12 before the control measures were enforced and decreased to 0.96 after the measures were implemented.

Full genome sequencing of the ANDV strain involved in this outbreak was performed with specimens from 27 patients and showed that the strain that was present (Epuyén/18–19) was similar to the causative strain (Epilink/96) in the first known Person-to-Person transmission of Hantavirus pulmonary syndrome caused by ANDV, which occurred in El Bolsón, Argentina, in 1996.

Clinical investigations involving patients with ANDV Hantavirus pulmonary syndrome in this outbreak revealed that patients with a High Viral Load and Liver Injury were more likely than other patients to spread infection.

Disease severity, genomic diversity, age, and time spent in the hospital had no clear association with secondary transmission.

Here is the seating plan of the Birthday Party held in Chubut Province, Argentina, that resulted in 34 confirmed infections and 11 Deaths.

Martínez and coworkers concluded that 6 of the 34 cases were exposed to Human Exhaled Droplet or Aerosol.

On the basis of both the epidemiologic and genomic investigations of Person-to-Person transmission events, it appears that Inhalation of Droplets or Aerosolized Virions may have been the routes of infection.

Andes Hantavirus Cytokine Storm

While many others have mentioned the Martínez et al. paper, I find the Supplementary file most interesting because they performed detailed analysis of Cytokines and other markers in the biological samples obtained from the living and the Dead.

This study aimed to present new data on the side effects of Favipiravir on healthy lung tissue and the respiratory system.

In the study, two different durations (5 and 10 days) were preferred to determine the effect of Favipiravir treatment due to clinical improvement rates of approximately 5 and 10 days during the use of Favipiravir in COVID-19 patients.

In addition, after 10 days of Favipiravir treatment, animals were kept for 5 days without any treatment to determine the regeneration of Lung tissues.

Favipiravir was administered to Rats by oral gavage at a daily dose of 200 mg/kg for 5 and 10 days, as in previous studies.

At the end of the experiment, the histopathological and biochemical effects of Favipiravir in the Lung tissue were investigated.

The data obtained from the study showed that Favipiravir increased oxidative stress parameters, expression of apoptotic markers, and pro-inflammatory markers in lung tissue.

Since Malondialdehydes is an oxidant parameter, it increased in Favipiravir-administered groups; It was determined that the antioxidant parameters glutathione, superoxide dismutase, glutathione peroxidase, and catalase decreased.

Other markers used in the analysis are Bcl-2, Bax, NF-κB, interleukin (IL)-6, Muc1, iNOS, P2X7R, IL-6 and caspase-3. The levels of Bax, caspase-3, NF-κB, IL-6, Muc1, and P2X7R were increased in the Fav-treated groups compared with the control. However, the levels of Bcl-2 decreased in the Fav-treated groups.

The present study proves that Favipiravir, widely used today, causes side effects in lung tissue.

Favipiravir is Genotoxic to Heart and Skin Cells

Favipiravir (T-705), used against influenza viruses, is approved for emergency use in many countries for the treatment of COVID-19.

The frequent adverse effects of Favipiravir are related with the gastrointestinal system, however, studies suggest a positive association of Favipiravir on QTc prolongation, which can cause cardiotoxicity.

Also, there are reports of Skin reactions such as Angioedema due to Favipiravir.

Despite the several adverse effects, studies examining the drug's effects at the molecular level are insufficient, e.g., the Genotoxic and oxidative stress-inducing effects of Favipiravir, which are among the primary mechanisms of drug-induced toxicity.

The cytotoxicity of Favipiravir was analyzed with the measurement of the ATP content in H9c2 cardiomyoblasts and CCD-1079Sk skin fibroblasts.

The ATP level decreased starting from 200 µM. The inhibitory effect on the Mitochondrial Electron Transport Chain enzymes complex I and complex V was also evaluated where Favipiravir showed significant enzyme inhibitory effects in the highest concentration studied.

A molecular docking study evaluating the interaction between Favipiravir-RTP and Mitochondrial DNA polymerase (POLG1) was done.

The relationship of Favipiravir with oxidative stress was examined by measuring Glutathione (GSH) and protein carbonyl levels which were observed higher after drug treatment compared to the control group.

The Genotoxicity study was done using the Comet assay and increase in DNA tail has been detected.

Furthermore, 8-OHdG levels were measured higher in Favipiravir treated cells indicating oxidative DNA damage.

Favipiravir induced oxidative stress leading to DNA damage in Cardiomyoblast cells and fibroblastic Skin cells.

Oxidative stress and DNA damage might eventually lead to organ-specific damage such as Cardiotoxicity and Dermal toxicity.

Considering the increased use of Favipiravir in recent years, and that oxidative stress and genotoxicity are two important indicators of drug-induced toxicity, the obtained results are worth attention.

Claims of value in Humans against Hantavirus

A 2013 paper, published by Toyama Chemical (a subsidiary of Fujifilm) the drug manufacturer reported that Favipiravir inhibited Hantavirus growth in vitro and daily dosing improved Hamster survival rates, but only if given before infection or before 3 days after infection.13 Toyama Chemical co-authors came from the Veterinary branch of the infamous US Rocky Mountain Regional Center of Excellence for Biodefense and Emerging Infectious Disease Research.

In 2022 Toshihiro Suzuki,14 Managing Director of YAKUGAI Ombudsperson of Medwatcher Japan demanded that Katsunobu Kato, then Minister of Health, Labour and Welfare, Stop Stockpiling Favipiravir (Avigan) And Revoke Its Approval.

The Favipiravir Deaths will result from a combination of direct drug killing via attack on the Liver, Kidneys and other vulnerable organs, its lack of efficacy against viruses and the reports of exacerbation of the disease.

Note “Foetal Exposure During Pregnancy” where the FDA does not include Dead Foetuses from Spontaneous Abortion as Deaths.

Poul Bak Thorsen has many co-authors in multiple countries and works with epidemiologists who find assocations of disease but his most useful works have involved correlation of measurable Cytokines and Genetic DNA mutations to explore causal links.

Mebendazole Tautomers, Desmotropes and Homosynthons

A little about the 3 types of Mebendazole before looking at Metabolites.

Tautomers are constitutional isomers of chemical compounds that rapidly interconvert through a process called tautomerization, which involves the migration of a Hydrogen atom (Proton) accompanied by a switch of a single bond and an adjacent double bond, often in dynamic equilibrium in solution. Precipitation of Mebendazole from various solvents yields dfferent ratios of tautomers.

Desmotropes are solid-state crystalline forms of a compound that represent different, isolable tautomeric structures.

Homosynthons are structural units in crystal engineering formed by the interaction of identical functional groups within a molecular crystal.

Please note the vital importance of Hydrogen Bonds linking Nitrogen atoms.

Found a nice 2024 paper by researchers in Argentina2 that helped me understand the previously mentioned A, B and C forms that differ widely in solubility, bioavailability and no doubt toxicity. The Supplementary file yielded the following 4 images.

Chiral Mebendazole Metabolite Derivatives

Liver Failure in Mebendazole dosed People was an issue linked to metabolites in 1982 by Australian researchers.3

In 1983 concern over Mebendazole Deaths prompted further research into the Human enzymes involved in Liver Failure but chirality was not considered.4

As mentioned earlier, Australian researchers corrected the record on the primary Mebendazole Metabolites in Humans in 1985.

They did not pay much attention to the chiral centres but warned that separate toxicology studies were needed before further Human trials.5

Subscribers will recall that in August 2022 I raised the Question6 of unknown toxicity of the Pfizer “novel” Lipid 4-hydroxybutyl azanediyl bishexane-6 1-diyl bis2-hexyldecanoate, known as ALC-0315, that contains chiral centres.7

In 2008 stereoselective formation of MBZ-OH in Humans, detected in plasma was reported by Swiss scientists.8

Abstract

An assay for the simultaneous determination of the enantiomers of hydroxymebendazole (OH-MBZ) and hydroxyaminoMebendazole (OH-AMBZ) together with aminoMebendazole (AMBZ) in human plasma is described for the first time.

It is based upon liquid-liquid extraction at alkaline pH from 0.5 mL plasma followed by analysis of the reconstituted extract by CE with reversed polarity in the presence of a 50 mM, pH 4.2 acetate buffer containing 15 mg/mL sulfated beta-CD as chiral selector.

For all compounds, detection limits are between 0.01 and 0.04 microgram/mL, and intraday and interday precisions evaluated from peak area ratios are <6.9 and <8.5%, respectively.

Analysis of 39 samples of echinoccocosis patients undergoing pharmacotherapy with Mebendazole (MBZ) revealed that the ketoreduction of MBZ and AMBZ is highly stereoselective.

One enantiomer of each metabolite (firstly detected peak in both cases) could only be detected.

The CE data revealed that OH-MBZ (mean: 0.715 microg/mL) is the major metabolite followed by AMBZ (mean: 0.165 microg/mL) and OH-AMBZ (mean: 0.055 microg/mL) whereas the MBZ plasma levels (mean: 0.096 microg/mL, levels determined by HPLC) were between those of AMBZ and OH-AMBZ.

In the text they state however that “traces of the other enantiomers, however, cannot be excluded” and “The assignment of the produced enantiomers could not be undertaken because the standards for the single enantiomers were not available”.

I will look for any subsequent publications by these authors.

This raises interesting questions that subscribers might like to discuss below.

In 2024 Italian researchers9 set out to learn more about Methyl (6-hydroxy(phenyl)methyl)-1H-benzo[d]imidazol-2-yl)carbamate) (MBZ-OH)

In healthy people MBZ-OH is mainly glucuronidated and excreted via the Human bile and is less poisonous to worms than MBZ. However people with Gilbert’s Disease have reduced activity of the Glucuronosyltransferase enzyme and therefore are more at risk of MBZ induced Liver Failure.10

Guglielmi et al. used HPLC and a special column to separate milligram quantities of the different optical isomers (enantiomers) of MBZ-OH.

As of August 2024, no studies of the genotoxicity of the enantiomers had been done.

Mebendazole Residues in Food

In 1991 German scientists used isolated perfused Rat Gut to study MBZ metabolites.11

In 1997 Dutch researchers12 looked at MBZ and its two main metabolites in the popular food, Eel.

Abstract

Mebendazole (MBZ) is extensively used in Eel culture for treatment of Pseudodactylogyrus spp. infections.

This use may lead to residues of MBZ in eel tissues.

Consequently, the residue profile of MBZ in Eel after treatment with the drug is of special concern. Therefore, a residue study was performed in European Eels (Anguilla anguilla), bath-treated with MBZ at a dose of 1 mg/liter for 24 hr and kept at a water temperature of 25 degrees C.

Liver, kidney, fat, skin, and muscle tissues samples were collected at intervals during and after treatment and analyzed for MBZ and its metabolites, hydroxy-MBZ (MBZ-OH) and amino-MBZ (MBZ-NH2), by HPLC.

Results showed that MBZ is extensively metabolized to MBZ-OH and MBZ-NH2. Liver and Kidney were found to contain the highest levels of MBZ metabolites, and Fat contained the highest levels of the parent compound.

Skin contained higher residue levels for all three compounds, compared with muscle tissue.

MBZ and its hydroxy metabolite were eliminated within 5 days from the edible parts (muscle and skin) of the eels, whereas MBZ-NH2 could be detected by the 14th day after the end of the treatment period.

Consequently, although MBZ and MBZ-OH constitute the residues of toxicological concern, MBZ-NH2 should be taken as the compound of interest for estimating the withdrawal time for consumption of eel treated with MBZ.

In 2001 Belgian researchers measured MBZ and metabolites in popular food Sheep Liver using Tandem Mass Spectrometry with detection limits ~ 1 μg/kg (1 ppb) but wth higher limits for inter-laboratory quantification.13

Maximum Residue Limits (MRLs) have been set for 2 MBZ metabolites, 5-hydroxymebendazole (HMBZ) and 2-amino-5- benzoylbenzimidazole (AMBZ), at 60 μg/kg (60 ppb) in South Korea.

With this in mind, Chinese researchers in Wuhan reported their MBZ and its metabolite residue measurements in 2018 in Bluntnose Black Bream, Shrimp, Eel and Turtle.14

In 2021 Chinese researchers published a high sensitivity method for analysis of MBZ residues in Chicken, Goose and Duck meat.15

A popular food with wealthier Japanese and some westerners is the Puffer Fish or Fugu. The level of MBZ and metabolites was measured in 2022 by Chinese researchers.16

Abstract

As a typical and broad-spectrum benzimidazole, Mebendazole (MBZ) has long been used in human and veterinary medicine to treat parasitic infestations, and is widely employed in the aquaculture of Japanese Pufferfish (Takifugu rubripes).

However, there have been no studies examining the pharmacokinetic characteristics of MBZ in Japanese pufferfish.

Furthermore, the presence of MBZ and its metabolites in animal-derived raw food represents a notable safety concern.

Here, we investigated the metabolism of MBZ using a UPLC-Q-TOF system. Additionally, we evaluated the pharmacokinetics of MBZ and two metabolites, 2-amino-5(6)-benzoylbenzimidazole (MBZ-NH2) and 5-hydroxymebendazole (MBZ-OH), in Japanese Pufferfish following intramuscular injection of 20 mg/kg MBZ. We detected three metabolites of MBZ (M1-M3), among which, 2-amino-5(6)-(a-hydroxybenzyl) benzimidazole (M3) was detected in an aquatic animal for the first time.

The plasma dispositions of MBZ, MBZ-NH2, and MBZ-OH were characterized by low plasma clearance, medium distribution volume, and long terminal half-life.

Moreover, these compounds were widely distributed in the muscle, from which they were rapidly cleared. The pharmacokinetics and metabolism of Mebendazole in Japanese Pufferfish are described for the first time in this study.

Our findings provide a basis for the rational application of MBZ in Japanese Pufferfish farming and contribute to our understanding of the metabolism of MBZ in cultured fish.

In 2022 researchers from Brazil reported measurements of MBZ in Hens Eggs but MRLs were not agreed despite may samples being considered “non-compliant”.17

Abstract

Analytical methodology for quantification of 15 antiparasitic drugs and their respective metabolites in laying hen eggs was optimized and validated.

The method uses acetonitrile as solvent extraction, sodium chloride for salting-out, low-temperature purification and analysis by LC-MS/MS.

A total of 348 egg samples were collected in 11 states of Brazil and 50% of the total samples presented antiparasitic residues, which were Albendazole, Fipronil, Fenbendazole, Ivermectin, Oxibendazole and Mebendazole.

A total of 12.4% of the samples were considered non-compliant, and residues quantified in these samples were Albendazole, Fipronil, and Mebendazole.

Albendazole was always identified as albendazole sulfone. Only one sample presented fipronil and fipronil sulfone; all others exclusively the sulfone metabolite. Fenbendazole was characterized by the presence of both metabolites: sulfone and sulfoxide.

Maximum Limits adopted are based on the Normative Instruction 51/2019 of the Brazilian Health Regulatory Agency (ANVISA), but Albendazole, Fipronil, Oxibendazole, Ivermectin, and Mebendazole do not have their Maximum Residue Level established.
